    Fluorescence-based proteasome activity profiling

    With the proteasome emerging as a therapeutic target for cancer treatment, accurate tools for monitoring proteasome (inhibitor) activity are in demand. In this chapter, we describe the synthesis and use of a fluorescent proteasome activity probe that allows for accurate profiling of proteasomal activity in cell lysates, intact cells, and murine and human patient-derived material, with high sensitivity using SDS-PAGE. The probe allows for direct scanning of the gel for fluorescent emission of the distinct proteasomal subunits and circumvents the use of Western blot analysis. Due to its suitable biochemical and biophysical properties, the fluorescent probe can also be used for confocal laser scanning microscopy and flow cytometry-based experiments

    . Banking at the present time is required to further improve its service,whereas customers are increasingly improving their mobility and needs. Servicein the banking industry is a very important thing in determining their success inthe face of competition. Quality of service is very influential on customersatisfaction and loyalty. The existence of decrease in the number of customers, adecrease in the amount of good incomes from savings, deposits and loans, also anincrease in the number of customers who complain significantly occur at thisprivate bank. This study was conducted to determine the effect of service qualityon customer satisfaction and loyalty on one Private Bank branches in CentralJakarta. The respondent in this study are the customers of the private bank inCentral Jakarta branch where the total number of customer at the private bankbranch in Central Jakarta are as many as 112 customers. The number ofrespondents in this study set out 88 respondents using a sampling technique calledpurposive sampling with slovin formula. This study uses analysis with the help ofthe SMART PLS program to determine the effect between service quality,customer loyalty and customer satisfaction, in addition, this study also uses thePearson r correlation with SPSS to determine the relationship between eachdimension. The results obtained from this study show that: 1) the quality ofservice gives positive and significant effect on customer satisfaction, 2) thecustomer satisfaction does not give positive and significant effect on customerloyalty, 3) the quality of service gives positive and significant effect on customerloyalty. The recommendation for this private bank in Central Jakarta branch is toimprove the quality of services provided to customers starting from theimprovement of the employee\u27s performance, system improvement, andtechnology improvement so that the customer is satisfied and loyal to this privatebank

    Electron transfer in photoexcited pyrrole dimers

    Following on from previous experimental and theoretical work [Neville et al., Nat. Commun. 7, 11357 (2016)], we report the results of a combined electronic structure theory and quantum dynamics study of the excited state dynamics of the pyrrole dimer following excitation to its first two excited states. Employing an exciton-based analysis of the Ã(π3s/σ*) and ˜ B (π3s/3p/σ*) states, we identify an excited-state electron transfer pathway involving the coupling of the Ã(π3s/σ*) and ˜ B (π3s/3p/σ*) states and driven by N–H dissociation in the ˜ B (π3s/3p/σ*) state. This electron transfer mechanism is found to be mediated by vibronic coupling of the ˜ B state, which has a mixed π3s/3p Rydberg character at the Franck-Condon point, to a high-lying charge transfer state of the πσ* character by the N–H stretch coordinate. Motivated by these results, quantum dynamics simulations of the excited-state dynamics of the pyrrole dimer are performed using the multiconfigurational time-dependent Hartree method and a newly developed model Hamiltonian. It is predicted that the newly identified electron transfer pathway will be open following excitation to both the Ã(π3s/σ*) and ˜ B (π3s/3p/σ*) states and may be the dominant relaxation pathway in the latter case

    Assessing the Relevance of Statistics and Crime Analysis Courses for Working Crime Analysts

    Crime analysts study criminal activity and police function to improve performance. Analysts inform operations, aid resource allocation, and evaluate programs. These tasks require high levels of statistical literacy. Given that most analysts are college-educated civilians, college coursework in statistics and/or crime analysis may represent the foundational knowledgebase within the profession. However, little research has attempted to determine if coursework teaches the skills needed by analysts. Underlying this issue is a limited understanding about what technical skills crime analysts regularly use. Analyzing parallel surveys of 98 criminal justice educators and 146 crime analysts, this study compares the skills taught in undergraduate-level statistics and crime analysis courses to those used by analysts. Comparisons are made between perceptions of the value of coursework. Results indicate discrepancies between the orientation of coursework and the needs of analysts which underscore differences in the perceived relevance of statistics education. Recommendations are discussed

    A Comparative Analysis of Potential Spatio-Temporal Access to Palliative Care Services in Two Canadian Provinces

    Background Access to health services such as palliative care is determined not only by health policy but a number of legacies linked to geography and settlement patterns. We use GIS to calculate potential spatio-temporal access to palliative care services. In addition, we combine qualitative data with spatial analysis to develop a unique mixed-methods approach. Methods Inpatient health care facilities with dedicated palliative care beds were sampled in two Canadian provinces: Newfoundland and Saskatchewan. We then calculated one-hour travel time catchments to palliative health services and extended the spatial model to integrate available beds as well as documented wait times. Results 26 facilities with dedicated palliative care beds in Newfoundland and 69 in Saskatchewan were identified. Spatial analysis of one-hour travel times and palliative beds per 100,000 population in each province showed distinctly different geographical patterns. In Saskatchewan, 96.7 % of the population living within a-1 h of drive to a designated palliative care bed. In Newfoundland, 93.2 % of the population aged 65+ were living within a-1 h of drive to a designated palliative care bed. However, when the relationship between wait time and bed availability was examined for each facility within these two provinces, the relationship was found to be weak in Newfoundland (R2 = 0.26) and virtually nonexistent in Saskatchewan (R2 = 0.01). Conclusions Our spatial analysis shows that when wait times are incorporated as a way to understand potential spatio-temporal access to dedicated palliative care beds, as opposed to spatial access alone, the picture of access changes
